Transformation of cytosine to uracil in single-stranded DNA via their oxime sulfonates.

چکیده
The novel transformation of cytosine oxime sulfonate (C*) to uracil sulfonate (U*) proceeded by a Co(II)-assisted benzoyl peroxide reaction, eventually leading to the conversion of cytosine to uracil.

AB Initio Calculations and IR Studies of Tautometric forms of Uracil and Cytosine and comparing results in different temperatures (25˚C, 37˚C and 40˚C).
In this paper, the molecular geometry for three tautomers of uracil and four tautomers of cytosine has been analyzed. vibrational IR spectra of the tautomers were investigated at HF and B3LYP level using the AB initio 6-31G* and LANL2DZ basis sets from the program package Gaussian 98 (A.7 Public Domain version). The physico-chemical and biochemical properties of uracil and cytosine are one of...
متن کاملNitric oxide-induced deamination of cytosine and guanine in deoxynucleosides and oligonucleotides.
The autoxidation of nitric oxide (NO.) forms the nitrosating agent N2O3, which can directly damage DNA by deamination of DNA bases following nitrosation of their primary amine functionalities. Within the G:C base pair, deamination results in the formation of xanthine and uracil, respectively. To determine the effect of DNA structure on the deamination of guanine and cytosine, the NO.-induced de...
متن کاملDNA N-glycosidases: properties of uracil-DNA glycosidase from Escherichia coli.
Uracil-DNA glycosidase, an enzyme that catalyzes the release of free uracil from uracil-containing DNA, has been purified ll,OOO-fold from Escherichia coli cell extracts. The enzyme preparation was essentially homogenous, as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The native enzyme is a small monomeric protein of a molecular weight of 24,500 + 1,000. The enzyme e...
متن کاملAn N-glycosidase from Escherichia coli that releases free uracil from DNA containing deaminated cytosine residues.
An enzyme that liberates uracil from single-stranded and double-stranded DNA containing deaminated cytosine residues and from deoxycytidylate-deoxyuridylate copolymers in the absence of Mg(++) has been purified 30-fold from cell extracts of E. coli. The enzyme does not release uracil from deoxyuridine, dUMP, uridine, or RNA, nor does it liberate the normally occurring pyrimidine bases, cytosine...
متن کاملAlternative induction of meiotic recombination from single-base lesions of DNA deaminases.
Meiotic recombination enhances genetic diversity as well as ensures proper segregation of homologous chromosomes, requiring Spo11-initiated double-strand breaks (DSBs). DNA deaminases act on regions of single-stranded DNA and deaminate cytosine to uracil (dU). In the immunoglobulin locus, this lesion will initiate point mutations, gene conversion, and DNA recombination. To begin to delineate th...
